Until 1957, McMaster University was controlled by the Baptist Convention of Ontario and Quebec. In 1957, it became a privately chartered, publicly funded, non-denominational institute. It was named McMaster after the Honourable William McMaster, a prominent Canadian Senator and banker who donated C$900,000 for the founding of the university.

The buildings at McMaster have all been built during different periods of time. For instance, the Hamilton Hall opened in 1926 whereas the university's nuclear research facility just newly opened in 2011. Plans to construct a new academic building in 2013, which will be known as the Wilson Building for Studies in Humanities and Social Sciences, are underway.
Infrastructure: Since 1928, McMaster University campus has constantly been undergoing development. The 6 original buildings on the main campus follow the neo-Gothic architectural style. They are now flanked by over 50 structures that were built predominantly during the 1950s, the 1960s, and the 2000s. The largest facility is the McMaster University Medical Centre, which is a multi-use research hospital that ranks amongst the largest paediatric academic health sciences centres in Canada and one of the busiest children's hospitals in Ontario. It is connected to the Life Sciences building and the Michael De Groote Centre for Learning & Discovery, which houses many well-funded research groups in areas of genetics, infectious diseases, and several specific conditions.
Since it began operation, the McMaster Nuclear Reactor (MNR) has been the largest university reactor in the Commonwealth. It is also the second-largest research reactor in the whole of North America. It is a "pool-type" reactor with a core of enriched uranium fuel that is moderated and cooled by distilled water. While the MNR is primarily used for research and the production of medical isotopes, the MNR also serves students in nuclear engineering, medical and health physics, and other applied radiation sciences.
McMaster University - Hamilton provides a wide range of irradiation, laboratory and holding facilities, including a cyclotron, an accelerator, a small-angle neutron-scattering detector, and wide-angle neutron scattering facilities. The cyclotron is used for the production of fluorine-18, which is used for research purposes; in particular, for the development of novel molecular imaging agents. The McMaster University Libraries include four libraries housing 1,645,876 volumes and 3,620,092 total resources, including videos, maps, sound recordings, and microfilm. Mills Library houses the humanities and social sciences collections, with a wide range of print and digital resources. Innis Library houses content which supports the academic and research interests of the DeGroote School of Business. Thode Library houses academic material of various disciplines of science and engineering. The Health Science Library houses books pertaining to medical sciences.
Faculties: McMaster University consists of six academic faculties, comprising Engineering, Health Science, Humanities, Social Sciences, Science, and the DeGroote School of Business. The Ron Joyce Centre at Burlington is home to DeGroote's MBA program and its business management program (both degree and non-degree programs). McMaster’s Downtown Centre located in Hamilton in the former Wentworth County Courthouse, is home to the McMaster Centre for Continuing Education which offers a variety of certificate and diploma programs as well as personal and professional development programs. The Michael G. DeGroote School of Medicine is located on the main campus as well as in two regional campuses at St. Catharines and Kitchener.
